Praying in the spirit is a unique aspect of our relationship with God that came into being after the gift of the Holy Spirit fell in Pentecost. So many times, however, we fail to utilize beauty and importance of this gift due to lack of understanding and incomplete teaching.
"For he who speaks in a tongue does not speak to men but to God, for no one understands him; however, in the spirit he speak mysteries...he who speaks in a tongue edifies himself...." 1 Corinthians 14: 3-4
Here, Paul tells us that when we pray in tongues, our spirits are praying. This prayer is the full, unaltered will of God for our lives. That why it's called a mystery; it's hidden from your natural mind and must be revealed by the Spirit. The reason why this prayer is perfect is because your spirit is responding to what it's receiving from the Holy Spirit, who lives in your innermost being.
The word Greek word for "edify" refers to a master builder. Praying in the spirit builds you up. What does it build you up into?
"But you, beloved, building yourselves up on your most holy faith, praying in the Holy Spirit." Jude 20 (emphasis mine)
When we pray in the spirit we are being built into a house of faith! The bible says in Hebrews that without faith it is impossible to please God. Praying in the spirit increases our capacity to please God. As we pray in the spirit, mysteries are released and declared, things that you wouldn't possibly think to ask for with your natural mind. Realize that you have a powerful destiny. It's so big that your mind isn't big enough to grasp it by just thinking about it, and since you can't grasp it you wouldn't be in the position to have faith for it. By praying in the spirit you pump yourself up to walk in a level of faith that you wouldn't be able to get on the strength of your own knowledge.

- There is a connection between the amount of time Paul prayed in tongues and his revelations
